From Superman: "Mission to San Caluma" by Jerry Siegel, Paul Cassidy and Paul Lauretta, Superman #6 (September 1940)
In this issue of Superman, there are moments where he seems to fly in a forward motion as opposed to simply "leaping over tall buildings". It's not quite conclusive as a first demonstration of the flight power, but we're moving in that direction, as evidenced by one of the stories showing him gliding using his cape.
Less flashy, but nonetheless of note, the issue also established Superman's microscopic vision and his blood's compatibility with four blood types!
From The Ray: "Origin of the Ray" by Will Eisner and Lou Fine, Smash Comics #14 (September 1940)
And over at Quality Comics, the Ray gets his first appearance, escaping from a weather balloon with new-found light powers, but still lacking gloves and a pair of pants! Somebody should say something. It's a little embarrassing.
